PROJECT 1: Analysis of Cryopreservation’s Effect on Cell Isolates and Manufactured Therapeutic Phenotypes

Test-Beds Involved: CAR-T & iPSC-CM

This project will determine whether cryopreservation alters the molecular profile or distribution of phenotypes within patient cell isolates or manufactured cell therapies that could affect bioprocessing parameters or clinical efficacy, respectively.

Aim 1: Determine the impact of cryopreservation on the molecular profile or phenotypical distribution of patient leukapheresis samples.

Aim 2: Determine the impact of cryopreservation on the molecular profile, activation state, or clinical efficacy of CAR-T cells.

Aim 3: Determine the effect of cryopreservation on cell viability, cell attachment, the molecular profile and functional activity of iPSC-CMs.

Aim 4: Develop novel reagents or culture approaches to counteract deleterious effects of cryopreservation on cell viability, phenotypical changes, or efficacy.
